Acute toxicity tests are short-term tests designed to measure the effects of toxic substances over the short life cycle of aquatic species. The acute toxicity test assessed the effect on survival within 24 to 96 hours. Fish are often used as model organisms for these tests because they have sensitive respiratory systems that make them highly susceptible to waterborne toxins. There are a variety of standardized testing methods available for determining the acute toxicity of chemicals to fish, each with its own advantages and limitations. Endpoints of Fish Acute Toxicity Test

Dose-response studies allow calculation of EC50, LC50, no observed effect concentration and lowest observed effect concentration for mortality.

Course of Fish Acute Toxicity Test

  • Acute toxicity tests are typically performed using five concentrations, a control, and a vehicle (i.e., solvent) control (if the vehicle is required), typically using 10 to 20 organisms per concentration.
  • Fish are exposed to the test substance for 96 hours by serial dilution. At the same time, a control test line is run without the test substance.
  • After 24, 48, 72, and 96 h, record mortality and test conditions (temperature, pH, and oxygen content).
  • The estimated LC50 (Lethal Concentration for 50% of the fish tested) is calculated from the test results based on the dose-response relationship. Based on the test data, LC0 (the highest concentration at which all fish survived) and LC100 (the lowest concentration at which all fish died) are also estimated.
